B7-1和B7-2及其受体CD28和CTLA-4构成了调节T细胞和B细胞反应的主要共刺激途径之一。尽管CTLA-4和CD28都可以与相同的配体结合,但CTLA-4与B7-1和B7-2的结合亲和力是CD28的20-100倍,并参与反应的下调。
B淋巴细胞活化抗原B7-1(简称B7)也称为分化簇80(CD80),是细胞表面球蛋白超家族的成员,在活化的B细胞、活化的T细胞、巨噬细胞和树突状细胞上表达。它是T细胞表面两种不同蛋白质的配体:CD28(用于自动调节和细胞间结合)和CTLA-4(用于减弱调节和细胞解离)。CD80与CD86协同工作以启动T细胞。CD80通过激活巨噬细胞中的NF-κB信号通路在诱导先天反应中发挥作用。因此,CD80被认为是自身性疾病和各种癌症的有希望的治疗靶点。
